For this reason, they are referred to as intrinsic chromophores. It is also possible chemically to attach arti cial groups with strong absorption spectra to proteins. Such groups are extrinsic chromophores and may be used as reporter groups because their spectra will be affected by their speci c environment. Ideally, reporter groups should have a single site of attachment to the target macromolecule and should not affect its normal structure and function. Absorption measurements are frequently the basis of detection methodologies in biochemistry. Such measurements may be made directly or indirectly. The variable wavelength detector used in chromatography systems mentioned above is a good example of direct spectroscopic detection (Section 3.3.2). Measurements at 210, 280 and 260 nm allow convenient on-line detection of peptides, proteins and DNA, respectively. Most biomolecules have relatively low intrinsic absorbance values, however, which limit their sensitivity in detection systems. Moreover, not every molecule in a complex mixture will have the same value for at a given wavelength. For these reasons, indirect measurements of a particular extrinsic chromophore with a strong absorption are widely used in detection systems. Examples include chromogenic substrates used in the detection of enzymes such as horseradish-peroxidase and -galactosidase (Table 3.3). These are nonbiological substrates with weak absorption spectra which are enzymatically converted into chromophores with strong spectra. Conjugation of such enzymes to speci c antibodies provides the basis for highly sensitive immunodetection in commercial diagnostic kits. High sensitivity is conferred by signal ampli cation, that is the production of many molecules of product by a single molecule of enzyme.
